Stratifying response to immunotherapy in NSCLC via cfDNA methylation and ISR transcripts

Identifying circulating biomarkers (cell-free DNA methylation + Integrated Stress Response transcripts) to predict response to immune checkpoint inhibitors in lung cancer. Goal: a non-invasive liquid biopsy to select the patients who truly benefit from immunotherapy.

Stratifying immunotherapy patients using circulating immune system–related transcripts and cfDNA methylation

Overall objective

Identification of transcriptional and post-transcriptional regulatory mechanisms of ISR genes related to immunotherapy response in advanced NSCLC patients. Prognostic stratification based on ISR transcript half-life and identification of predictive biomarkers for acquired resistance.

Expected results

Successful stratification of advanced NSCLC patients based on the regulation of ISR transcripts and their half-life dynamics. Identification of novel transcriptional and post-transcriptional mechanisms that correlate with immunotherapy resistance or response. Discovery of predictive biomarker signatures that can be used to forecast acquired resistance to immunotherapy. Improved understanding of the interaction between ISR gene expression and clinical outcomes, leading to better prognostic tools.
